[68Ga]Ga-DOTA-H2D3 is an investigational radiopharmaceutical immunoPET imaging probe developed by RenJi Hospital. It consists of a HER2-targeting antibody fragment (H2D3) conjugated to the macrocyclic chelator DOTA and radiolabeled with the positron-emitting isotope Gallium-68 (68Ga). The agent is designed to bind specifically to the Human Epidermal Growth Factor Receptor 2 (HER2), which is frequently overexpressed in various malignancies, including gastric and other digestive system carcinomas. By utilizing positron emission tomography (PET) imaging, [68Ga]Ga-DOTA-H2D3 allows for the non-invasive assessment of HER2 expression levels and the detection of metastatic lesions, potentially aiding in patient stratification for HER2-targeted therapies. It is currently being evaluated in clinical trials for the diagnostic imaging of digestive system carcinomas.
